Vaccinia virus OPG111 유전자를 E.coli에서 발현시킨 재조합 단백질이다. 본 효소는 supercoiled DNA의 한 가닥을 끊어 DNA의 supercoiling을 감소하도록 DNA가 회전하게 한 후 끊었던 부분을 다시 봉합함으로써, DNA에 작용하는 비틀림 응력을 감소시킨다. Positive, Negative supercoiling 두 가지 모두 완화시킬 수 있다. (C/T)CCTT 서열의 3’쪽 단일가닥 nick을 생성한다. 다른 DNA 또는 RNA가 nick이 생성된 반 대편 가닥에 적절하게 결합한다면 해당 위치에서 DNA 재조 합이 일어날 수 있다. 낮은 농도(2.5 mM)의 Mg2+은 효소의 활성을 증가시킬 수 있다.
